Iovance Biotherapeutics posted third-quarter 2025 product revenue of about $68 million, up 13% sequentially, with U.S. sales of its TIL therapy Amtagvi contributing roughly $58 million and Proleukin about $10 million. Gross margin improved to 43% as cost-optimization efforts took hold, and the company reaffirmed full-year 2025 revenue guidance of $250 to $300 million. The U.S. launch infrastructure now spans more than 80 authorized treatment centers across nearly 40 states, community sites have begun treating patients, the average manufacturing turnaround is 32 days, and a specialty pharmacy agreement with InspiroGene by McKesson aims to broaden access. Internationally, Health Canada approved Amtagvi in August, with potential decisions in the United Kingdom and Australia in the first half of 2026 and in Switzerland in 2027, while a European regulatory strategy is being finalized. On the pipeline, interim data in nonsquamous NSCLC showed a 26% objective response rate and a median duration of response not reached beyond 25 months, supporting a potential sBLA and a 2027 launch if timelines hold.
The strategic question is whether Iovance can convert early momentum into a durable cell therapy franchise by scaling manufacturing, compressing cycle times, and proving value to payers outside of academic centers. TIL therapy is no longer a scientific curiosity; it is a commercial product with a complex, time-sensitive workflow spanning tumor harvest, bespoke manufacturing, and coordinated inpatient and outpatient care. The move to centralize manufacturing at the Iovance Cell Therapy Center in 2026 is designed to improve margins and reduce external costs. Still, success will hinge on execution in real-world settings, where referral patterns, scheduling bottlenecks, and supportive care logistics can make or break throughput.
For patients, the community-site activation is pivotal, moving access closer to where care is delivered and reducing reliance on a handful of academic hubs. For payers, the story will be durability and total cost of care. A 32-day vein-to-ship average is notable, but the economic argument improves only if real-world outcomes mirror trial-grade durability and if complications are contained through standardized protocols and improved IL-2 regimens. Medical Affairs teams will need to operationalize evidence quickly: targeted education on patient selection, adverse event management, and site readiness; prospective RWE to validate durability and resource utilization; and clear communication around potency assays and comparability as manufacturing scales.
Competition is intensifying, and Iovance is trying to turn its first-mover advantage in solid-tumor cell therapy into a platform position as the field diversifies. Gene-edited TILs (IOV-4001), modified IL-2 backbones (IOV-3001), and IL-12-armed constructs (IOV-5001) reflect a broader industry shift toward engineering for potency, persistence, and tolerability. If the NSCLC study reads out positively and the sBLA stays on track, TILs could expand from post-PD-1 melanoma into larger solid-tumor markets, intensifying comparisons with antibody-drug conjugates, bispecifics, and next-line immunotherapy combinations. Global expansion is another test: Canada’s nod is encouraging, but EU pricing, ATMP infrastructure, and country-by-country assessments will determine whether an addressable population measured in tens of thousands becomes a commercially meaningful footprint.
The following two years will clarify whether manufacturing centralization, community-site integration, and stronger RWE can lift margins while accelerating adoption. If NSCLC data sustain their durability signal and logistics improve toward a sub-month cycle time, does TIL therapy become a standard second-line option across multiple tumors—or does payer pressure and operational friction keep it a niche reserved for the most motivated centers and patients?
